Flexuous rod particles of 11 × 800 nm were isolated from cucumber (Cucumis sativus L.) from a commercial hydroponic farm in Sembawang, Singapore, in 1989. Affected plants showed leaf yellow mosaic, severe fruit distortion, and stunting. Both flexuous rods and cytoplasmic inclusion protein were observed by transmission electron microscopy in crude leaf sap from diseased plants. The virus was isolated and identified by electron microscopy as zucchini yellow mosaic virus (ZYMV). Host ranges included Citrullus lanatus (Thunb.) Matsum. & Nakai, Cucumis melo L., C. sativus, Cucurbita pepo L., Luffa acutangula (L.) Roxb., and L. cylindrica Roxb. The isolate did not infect Momordica charantia L. or Trichosanthes anguina L. Results of indirect ELISA were positive when the mosaic cucumber leaves were tested with known ZYMV virions and ZYMV cytoplasmic inclusion protein antisera. This is the first report of ZYMV from Singapore.
