Job Details
A two-year postdoctoral position is available starting in April 2013 to examine the pathogenesis of grapevines by the wood-infecting fungus Neofusicoccum parvum, which causes Botryosphaeria canker. The objective of this project is to define the stages of canker development with the ultimate goal of developing an early-detection tool, which would greatly improve control of this disease in the field. We are using the combination of RNA-seq to examine the grape transcriptomic response to infection and a novel CT scan-type method at the Advanced Light Source (i.e., the synchrotron at the Lawrence Berkeley National Lab) for imaging canker development and damage to the vascular system – high-resolution computed tomography (HRCT). Colonization of woody tissues by vascular pathogens is traditionally examined by sectioning and microscopy, but this approach is destructive and it limits investigation to cells within the sections. HRCT provides a more comprehensive examination of the internal vasculature because it allows for investigation of the intact woody stem of a live plant. Additional beamlines at the Lawrence Berkeley National Lab provide even greater resolution and detect wavelength profiles of various plant and fungal compounds, and may be additional sources of complimentary data. In addition to the Baumgartner lab, the project includes two other labs: Dr. Andrew McElrone (USDA, Agricultural Research Service, Davis, CA – responsible for examining the plant-anatomical response in the grape wood, using HRCT) and Dr. Grant Cramer (University of Nevada, Reno – responsible for examining the plant-transcriptomic response in the leaves, using RNA-seq). The Baumgartner lab is responsible for designing all greenhouse experiments, gathering and cataloging samples, and using culture, microscopic, and/or nucleic acid-based methods to track the stages of infection. Additionally, the postdoc will work very closely with our collaborators to decide which samples are to be sent for RNA-seq and how to interpret the results of the analyses. As such, knowledge of how plants respond to infection (at the transcriptome level, specifically) in various plant-fungal pathosystems is essential. The research is part of a large project funded by the USDA-NIFA Specialty Crop Research Initiative, with the long-term goals of developing detection, research, and extension tools for wood-canker diseases of grape, pistachio, and almond. Research will be conducted at the Department of Plant Pathology, University of California, Davis.
Job Requirements
A recent Ph.D. (within one year of completion) in plant pathology, microbiology, mycology, genetics, or a related scientific discipline is required. Applicant must have experience with genomic techniques that evaluate a plant’s response to pathogen infection, experience with light and/or confocal microscopy, and skill in conducting greenhouse experiments using appropriate experimental design and statistical analyses.
